ADAM10 is a member of the ADAM (a disintegrin and metalloprotease like domain) family.
It cleaves the membrane-bound precursor of TNF-alpha at 76-Ala-|-Val-77 to its mature soluble form and is responsible for the proteolytic release of several cell-surface proteins, including heparin binding epidermal growth-like factor, ephrin-A2 and for constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP).
It contributes to the normal cleavage of the cellular prion protein and is involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicules, suggesting a vesicle-based protease activity.
Applications:Suitable for use in Direct ELISA, Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Direct ELISA: Shows ~20% crossreactivity with recombinant mouse ADAM10, ~5% crossreactivity with recombinant human TACE and <1% crossreactivity with recombinant human ADAM8, ADAM9, ADAM15 and BACE
Western Blot: 1ug/ml under non-reducing conditions.
Immunohistochemistry: 5-15ug/ml with immersion fixed paraffin-embedded sections of human prostate cancer tissue
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months after receipt at -20°C.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
