Presenilin 1 (PS1) and Presenilin 2 (PS2) are catalytic subunits of the gamma-secretase complex, which is an endoprotease complex that catalyzes the intramembrane cleavage of integral membrane proteins such as Notch receptors and APP (beta-amyloid precursor protein).
The other members of the gamma-secretase complex are required to have a protease activity.
PS1 and PS2 may play a role in intracellular signaling, gene expression and in linking of chromatin to nuclear membrane.
It regulates epithelial-cadherin function.
Three causative genes have been identified.
Mutation of these genes lead to presenile Alzheimer's disease.
They are APP (amyloid precursor protein gene), PSEN1 and PSEN2.
The PSEN2 gene provides instructions for making a protein named presenilin 2.
Presenilin 2 processes certain proteins that are important for transmitting biochemical signals from the cell membrane into the nucleus.
In the nucleus, these signals activate particular genes that are important for cell growth and maturation.
