53BP1 binds to the tumor suppressor protein p53 and has a potential role in DNA damage responses.
Small interfering RNA (siRNA) directed against 53BP1 in mammalian cells to demonstrated that 53BP1 is a key transducer of the DNA damage checkpoint signal.
53BP1 was required for p53 accumulation, G2-M checkpoint arrest, and the intra-S-phase checkpoint in response to ionizing radiation.
53BP1 played a partially redundant role in phosphorylation of the downstream checkpoint effector proteins Brca1 and Chk2 but was required for the formation of Brca1 foci in a hierarchical branched pathway for the recruitment of repair and signaling proteins to sites of DNA damage.
Applications:Suitable for use in Immunofluorescence, Western Blot and Immunocytochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: a band is seen at ~220kD
Immunofluorescence: 1:500.
Immunofluorescent staining of mouse B cells.
The cells were first fixed with Formaldehyde and permeabilized using Triton-X100.
The 53BP1 antibody was incubated for one hour at room temperature in PBS + 0.2% BSA + 0.1% Tween 20.
The secondary antibody used was an Alexa fluor 633 goat anti-rabbit used at a dilution of 1:500, and was incubated at RT for one hour in PBS, 0.2% BSA, 0.1% Tween 20
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
