• TBX6 (T-box protein 6) is a 47kD member of the T-box family of transcriptional regulators.
• It is expressed during embryogenesis, plays a role in presomitic mesoderm development, and promotes delta-like 1 transcription.
• Human TBX6 is 436 amino acids (aa) in length.
• It contains one T-box DNA-binding domain (aa90-277).
• There is one alternate splice form that shows a 15 aa substitution for aa 281-436.
• Over aa 1-280, human TBX6 shows 94% and 90% aa identity with canine and mouse TBX6, respectively.
• Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
• Other applications not tested.
• Recommended Dilution:Direct ELISA: 0.5-1ug/ml.
• The detection limit is ~2ng/well
• Western Blot: 1ug/mlImmunohistochemistry (fluorescence): 10ug/ml detected T-box 6 in cryosections of E10.5 mouse embryos.
• Tissue was fixed overnight in buffered zinc formalin fixative at 4ºC.
• Sections were blocked with 10% donkey serum, 0.1% Triton in 1% BSA in PBS for 45 minutes at RT.
• After blocking sections were incubated with diluted T1848-30 for 3 hours at RT folled by a Dk xGt IgG (NorthernLights 557) at RT for 1 hour.
• Between each step, sections were washed with 0.1% BSA, PBS
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized powder may be stored at -20°C.
• Stable for 12 months at -20°C.
• Reconstitute with sterile PBS.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
