Human prostate specific membrane antigen (PSMA), a tumor marker in prostate cancer encoded by the FOLH1 gene, is a type II transmembrane zinc metallopeptidase that is most highly expressed in the nervous system, prostate, kidney, and small intestine (1, 2).
The enzyme is also known as glutamate carboxypeptidase II (GCPII), folate hydrolase 1, folypoly-gamma-glutamate carboxypeptidase (FGCP), and N-acetylated-alpha-linked acidic dipeptidase I (NAALADase I).
In the brain, PSMA hydrolyzes the neurotransmitter N-acetyl-Asp-Glu to produce glutamate, another neurotransmitter.
Inhibition of brain PSMA activity is considered to be a promising approach for the treatment of neurological disorders associated with glutamate excitotoxicity, such as stroke, chronic pain, and amyotrophic lateral sclerosis (3).
Intestinal PSMA hydrolyzes folylpoly-gamma-lutamates, facilitating the uptake of folate (4).
Applications:Suitable for use in ELISA, Western Blot, Flow Cytometry and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Direct ELISA: 0.5-1.0ug/ml.
The detection limit is ~0.2ng/well
Western Blot: 0.1-0.2ug/ml.
The detection limit is ~5ng/lane under non-reducing and reducing conditions
Flow Cytometry: 25ug/ml.
10ul labeles 1-5x10e5cells in a total reaction volume ≤200ul.
Tested using LnCap cells
Immunohistochemistry: 2-5ug/ml detects PMCA in cells and tissues

Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months at -20°C.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
