• PEAR1 (platelet endothelial aggregation receptor 1; also MEGF12) is a 150kD member of the MEGF family of proteins.
• It is expressed principally on platelets and endothelium, and undergoes phosphorylation in response to platelet activation and adhesion.
• Mature human PEAR1 is a type I transmembrane protein that is 1017 amino acids (aa) in length.
• It contains a 735 aa extracellular region (aa 21-755) and a Pro-rich 261 aa cytoplasmic domain.
• The extracellular region shows one EMI segment (aa 25-103) followed by nine EGF-like repeats (aa 104-692).
• There is one potential alternate start site at Met376.
• Applications:Suitable for use in Direct ELISA, Western Blot and Flow Cytometry.
• Other applications not tested.
• Recommended Dilution:Flow Cytometry: 10ug/ml.
• Use 10ul to label 1-5x10e5 blood derived CD41+ platelets.
• Direct ELISA: 0.5-1.0ug/ml.
• Detection limit for rhPEAR1 is~ 0.3ng/well.
• Western Blot: 0.1-0.2ug/ml.
• Detection limit for rhPEAR1 is ~ 1ng/lane and 10 ng/lane under non-reducing and reducing conditions, respectively.
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized powder may be stored at -20°C.
• Stable for 12 months at -20°C.
• Reconstitute with sterile PBS.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
