Insulin-like Growth Factor Binding Protein-Like 1 (also IGFBP-rP4) is a secreted member of the IGFBP family of proteins.
Based on human, mature mouse IGFBP-L1 is a 251 amino acid (aa), 38kD glycoprotein.
It contains an N-terminal IGFBP motif (aa 53-68), a kazal-type serine protease inhibitor region (aa 83-143) and a C-terminal Ig-like domain (aa 161-237).
IGFBP-L1 is expressed in brain and testis and may be a tumor suppressor, possibly through the induction of apoptosis.
Over aa 18-270, mouse IGFBP-L1 shares 96% and 79% sequence identity with rat and human IGFBP-L1, respectively.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-0.2ug/mL detection limit for rmIGFBP-L1 ~ 2ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively
Immunohistochemistry: 10ug/mL detects IGFBP-L1 in cryosections of E13.5 mouse brain.
Tissue was fixed overnight in 4% PFA plus14% picric acid in PBS at 4° C.
Sections were blocked with 10% donkey serum and 0.1% Triton in 1% BSA in PBS for 45 minutes at RT.
After blocking, sections were incubated with diluted primary antibody for 3 hours at RT followed by dye-conjugated streptavidin at RT for 1 hour.
Between each step, cells were washed with PBS containing 0.1% BSA.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months at -20°C.
Reconstitute with sterile ddH2O or PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
