• Recommended Dilutions:Western Blot: ≥1:500 Immunohistochemistry: 1:400-1:800 in PBS/0.3% Triton X-100, Cy3 technique; 1:5000-1:10,000 in PBS/0.3% Triton X-100, Biotin/Avidin-HRPOptimal dilutions to be determined by the researcher.
• Immunohistochemistry Application Notes:Perfusion Fixation: Fixative- 4% paraformaldehyde in 0.1M phosphate buffer, pH 7.4; 500ml, over 20 minutes
• Post Fixation: 4% paraformaldehyde in 0.1M phosphate buffer, pH 7.4; 500ml, over 20 minutes
• Post Fixation: 1.5 hour at 4°C.
• 4% paraformaldehyde in 0.1M phosphate buffer, pH 7.4
• Note: Paraformaldehyde is a necessary component in fixation of serotonin.
• If needed, low levels of glutaraldehyde (0.1- 0.3%) may be used in conjunction with paraformaldehyde
• Sections: 10um cryostat or 50um vibratomeAbsorption control: 5-HT/BSA 25ug/ml diluted serum.
• Incubation on Tissue: 16-24 hours at 4°C.
• Positive Control:Rat hypothalamus, raphe nuclei, spinal cord
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
• Reconstitute with sterile TBS or PBS.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
