This gene is a member of the leucine-rich repeat kinase family and encodes a protein with an ankryin repeat region, a leucine-rich repeat (LRR) domain, a kinase domain, a DFG-like motif, a RAS domain, a GTPase domain, a MLK-like domain, and a WD40 domain.
The protein is present largely in the cytoplasm but also associates with the mitochondrial outer membrane.
Mutations in this gene have been associated with Parkinson disease-8.
Applications:Suitable for use in ELISA, Immunohistochemistry (Paraffin), Immunoflourescence and Western Blot.
Other applications not tested.
Recommended Dilution:Immunohistochemistry: 1:100 Immunofluorescence: 1:1000 (Mouse CAD cells transfected with human wild-type LRRK-2, immunoprecipitation: 7ul/15 cm plate of SH-SY5Y cells grown to 80% conflue, Western Blot: 1:1(the theoretical molecular weight of LRRK2 is ~286kD.
Other bands have been seen with some lysates, but these bands have been blocked by the control peptide, L4502-21E, suggesting that these bands are degredation products)Optimal dilution determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
