Stabilin-2, also known as HARE (hyaluronan receptor for endocytosis) and FEEL-2, is a type I transmembrane multi-domain protein that is the closest homolog of Stabilin-1.
Its large extracellular domain contains seven fasciclin, multiple EGF-like, laminin type EGF-like, and a link domain.
The short cytoplasmic tail of Stabilin-2 lacks the CGA interactive motif found in Stabilin-1.
Instead, it contains a motif for the classical cargo adaptor, AP2.
Stabilin-2 is a scavenger receptor that is expressed in the sinusoidal endothelial cells of liver, spleen, and lymph nodes.
It binds and mediates the systemic clearance of hyaluronan, AGE, collagen N-terminal propeptides and acetylated LDL.
By proteolytic cleavage, two isoforms (315 kD and 190 kD) with similar activities can be generated.
Over the region used as immunogen, human and mouse Stabilin-2 share 86% amino acid sequence identity.
Applications:Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human Stabilin-2.
The detection limit for rhStabilin-2 is approximately 10 ng/lane under non-reducing and reducing conditions.
In this format, this antibody shows less than 1% cross-reactivity with rhStabilin-1.
Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
