Semaphorin 3E (previously SemaH) is a secreted protein belonging to the semaphorin family.
Semaphorin 3E is highly expressed by a subset of motor neurons in developing somites, where it acts as a repulsive cue for PlexinD1-expressing endothelial cells of adjacent intersomitic vessels.
Like other Semaphorin 3 proteins, the 775 aa human Semaphorin 3E contains a signal sequence, a Semaphorin domain, a C2-like Ig domain and a C-terminal basic domain.
Disulfide-linked dimerization and furin-type proteolytic processing increase Semaphorin 3E bioactivity.
Mature human and mouse Semaphorin 3E share 90% aa identity.
Applications:Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 0.1-0.2ug/ml with the appropriate secondary reagents to detect human Semaphorin 3E.
The detection limit for rhSemaphorin 3E is approximately 5ng/lane under non-reducing and reducing conditions.
Immunocytochemistry: 10ug/ml to detect Semaphorin 3E on MDA-MB-453 human breast cancer cells.
Immunohistochemistry: 10ug/ml to detect Semaphorin 3E in cryosections of E13.5 mouse embryos
Direct ELISA: 0.5-1.0ug/ml with the appropriate secondary reagents to detect human Semaphorin 3E.
The detection limit for rhSemaphorin 3E is approximately 1.0ng/well.
In this format, this antibody shows approximately 70% cross-reactivity with rmSemaphorin 3E and less than 5% cross-reactivity with rmSemaphorin 3C and rmSemaphorin 3F.
