Versican (versatile proteoglycan; also known as PG-M) is a 500-600kD, secreted member of the Aggrecan/Versican proteoglycan family.
It is a variably glycanated, hyaluronan-binding, chondroitin sulfate-containing molecule that likely serves an anti-adhesion function and regulates the hydration properties of pericellular matrices.
Human Versican is 3396 amino acids (aa) in length.
It contains an N-terminal hyaluronan-binding region (G1; aa21-346), two large, alternately spliced GAG attachment domains (GAGalpha; aa348-1335; GAGbeta aa1336-3088) and a C-terminal selectin-like region (G3; aa3089-3396).
Over aa1344-1554, human Versican shares 80% and 60% aa sequence identity with canine and mouse Versican, respectively.
Applications:Suitable for use in Direct ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 0.1ug/ml detects Versican in recombinant human Versican isoform V0.
Immunocytochemistry: 5-15ug/ml detects Versican in immersion fixed HeLa cells.
Immunohistochemistry: 5-15ug/ml detects Versican in immersion fixed frozen sections of mouse embroyo.
Optimal dilutions should be determined by each laboratory.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
