• Interferon regulatory factor 3 (IRF3) is a 50kD, 427aa, member of the IRF family of proteins.
• Human IRF3 is a transcription factor that binds the interferon-sensitive response element (ISRE) and is activated by Toll-like receptors, TLR3 and TLR4.
• Viral entry or engagement of TLR3 or TLR4 stimulates IRF3 phosphorylation, nuclear translocation and stimulation of IFN production.
• Over the region used as immunogen, aa206-427, human IRF3 is 76% and 83% identical to the corresponding mouse and porcine proteins, respectively.
• Applications:Suitable for use in Western Blot, Flow Cytometry and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilutions:Western Blot: 1ug/ml in lysates of HeLa human cervical epithelial carcinoma cell line, U937 human histiocytic lymphoma cell line, Raji human Burkitt's lymphoma cell line, and Daudi human Burkitt's lymphoma cell lineFlow Cytometry: 2.5ug labels 10e6 cells in Daudi Human Cell LineImmunocytochemistry: 5-15ug/ml detects human IRF3 in HeLa cells.
• Cells were fixed with PBS, 4% paraformaldehyde for 20 min at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 min at RT.
• After blocking, cells were incubated with diluted primary antibody overnight at 4°C followed by fluorochrome-coupled anti-goat IgG at RT in the dark for one hour.
• Between each step, cells were washed with PBS, 0.1% BSA.
• Knockout Validated: IRF3 is specifically detected in HeLa human cervical epithelial carcinoma parental cell line but is not detectable in IRF3 knockout HeLa cell line
• Optimal dilutions to be determined by researcher.
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
• Reconstitute with sterile PBS.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
