HSF4 (heat shock factor 4) is a 55 kD member of the HSF family of transcription factors.
It is highly expressed in lens where it maintains lens integrity and blocks FGF synthesis.
Mouse HSF4B is 492 amino acids in length.
It contains one DNA binding region (aa # 17-122) and a Leu-zipper trimerization domain (hydrophobic repeat-A/B) (aa # 129-203).
Two additional potential isoforms exist.
The first isoform, termed HSF4A, is transcriptionally inactive and shows a 45 aa substitution for aa # 245-319.
The second isoform shows an 18 aa substitution for the first 78 amino acids of the N-terminus.
Over aa # 343-492, mouse HSF4 shares 93% and 72% aa sequence identity with rat and human HSF4, respectively.
Applications:Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse HSF4.
The detection limit for rmHSF4 is approximately 1 ng/lane under non-reducing and reducing conditions.
In this format, this antibody shows approximately 25% cross-reactivity with rhHSF4 and less than 5% cross-reactivity with rmHSF1 and rhHSF2.
This antibody has not been optimized for use in Western blot with cell lysates.
Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse HSF4.
The detection limit for rmHSF4 is approximately 0.2 ng/well
Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
