Platelet glycoprotein Ib alpha chain (GPIb-alpha), also known as CD42b alpha, is a 145kD platelet type I transmembrane protein.
It binds von Willebrand factor (VWF) and thrombin, and is part of the platelet GPIb-IX-V complex, which plays important roles in vascular biology, including hemostasis and thrombosis.
GPIb-alpha is also a member of the family of proteins with leucine-rich repeats, which mediate protein-protein interactions.
The 610 residue mature human GPIb-alpha contains a 489aa extracellular domain (ECD) (aa17-505) and a 100aa cytoplasmic region (aa527-626).
Proteolytic cleavage of the GPIb-alpha ECD from platelet surface releases soluble GPIb-alpha (glycocalicin), which is found in plasma.
The ECDs of human, mouse and canine GPIb-Alpha share approximately 50% aa sequence identity.
Applications:Suitable for use in Flow Cytometry, Western Blot and ELISA.
Other applications not tested.
Recommended Dilution:Flow Cytometry: 25ug/ml; Add 10ul of diluted antibody to 1-2.5x10e5 cells in a total reaction volume no greater than 200ul.
Tested using blood-derived CD41+ platelets
Western Blot: 0.1-0.2ug/ml; Detection limit under non-reducing and reducing conditions of rhGPIb-alpha is ~1ng/lane
ELISA: 0.5-1ug/ml; Detection limit of rhGPIb-alpha is ~5ng/well
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months at -20°C.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
