Type I insulin-like growth factor receptor (IGF-IR) is a transmembrane tyrosine kinase.
It is widely expressed among many cell types in fetal and postnatal tissues and many cell lines.
Upon binding of its ligands, IGF-I and IGF-II, autophosphorylation of receptors occurs.
The triple tyrosine cluster (Tyr1131, Tyr1135 and Tyr1136) within the kinase domain is the earliest major site of autophosphorylation.
Phosphorylation of these three tyrosine residues is necessary for activation of kinases.
Insulin receptors (IRs) share significant similarity with IGF-I receptors in structure and function.
There is also an equivalent triple tyrosine cluster (Tyr1146, Tyr1150 and Tyr1151) in its activation loop of the kinase domain.
Tyrosine autophosphorylation of the receptors is one of the earliest cellular responses to insulin stimulation.
The autophosphorylation begins by phosphorylation of Tyr1146 and either Tyr1150 or Tyr1151.
The full activation of receptor kinase activity depends on the triple tyrosine phosphorylation.
Applications:Suitable for use in Western Blot.

Recommended Dilution:Western Blot: 1:1000 detected phospho-IR (Tyr1158/Tyr1162/Tyr1163) in RIPA lysate of untreated and insulin treated CHO cells transfected with IR

Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
