• Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts.
• TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions.
• Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis.
• Applications:Suitable for use in ELISA and Western Blot.
• Other applications not tested.
• Recommended Dilution:ELISA: To detect mTNF-alpha by direct ELISA (using 100ul/well antibody solution) this antibody can be used at a concentration of 0.15-0.30ug/ml.
• Used in conjunction with compatible secondary reagents, allows the detection of at least 0.2ng/well of recombinant mTNF-alpha
• Western Blot: To detect mTNF-alpha by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml.
• Used in conjunction with compatible secondary reagents the detection limit for recombinant mTNF-alpha is 1.5-3.0ng/lane, under either reducing or non-reducing conditions
• Optimal dilutions to be determined by the researcher.
• Reconstitution:Reconstitute to a concentration of 50ug/ml with sterile PBS solution containing 0.1% BSA, 40-50% glycerol.
• Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
• Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
