Human Bone Morphogenetic Protein 2 (BMP2) is a disulfide-bonded homodimeric protein with an apparent molecular weight of 26kD.
BMP2 regulates similiarly to its nearest homologue BMP-4 diverse fundamental processes during embryonic development: BMP2 and other BMP proteins have great potential for medical therapeutic applications, in particular because they allow or at least accelerate the ossification of extensive bone lesions.
BMP2 lacks the natural N-terminus which results in a 15-20 fold increase of specific activity.
BMP2 is a heparin binding protein.
Applications:Suitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization.
Other applications not tested.
Recommended Dilution:ELISA (Sandwich): 0.5-2ug/ml (100ul/well); Used in conjunction with 208604 BMP-2 (Biotin) detects 0.2-0.4ng/well of recombinant hBMP-2 Western Blot: 0.1-0.2ug/ml; Detection limit is 1.5-3.0ng/lane for recombinant hBMP-2 under either reducing or non-reducing conditions
Neutralization: 1.3-1.9ug/ml; Yields one-half maximal inhibition [ND50] of the biological activity of hBMP-2 (200ng/ml)
Immunohistochemistry: 0.25ug/ml Formalin fixed paraffin embedded human colon/rectum adenocarcinoma with an overnight incubation at 4°C Requires heat induced antigen retrieval with a pH 6.0 sodium citrate buffer.
An HRP-labeled polymer detection system was used with a DAB chromogen.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
