• Mouse Calgranulin A, also known as CP-10, S100A8 and MRP8, is a 10kD member of the S100 family of calcium- binding proteins.
• S100A8 contains short sequential modules, including an N-terminal alpha-helix, a Ca++-binding EF-hand segment, a short central linker region, a second EF-hand segment and a C-terminal alpha-helix.
• S100A8 noncovalently heterodimerizes with S100A9.
• In the presence of Ca++, the heterodimers form heterotetramers.
• The dimeric complex is found both intracellularly and extracellularly.
• It binds to heparan sulfate and is chemotactic for PMNs and macrophages.
• The amino acid sequence of mouse S100A8 is 80% and 57% identical to that of rat and human S100A8, respectively.
• Applications:Suitable for use in Western Blot, Immunocytochemistry and ELISA.
• Other applications have not been tested.
• Recommended Dilutions:Western Blot: 0.25ug/ml using lysates of mouse spleen tissueImmunocytochemistry: 5-15ug/ml detects S100A8 in mouse NmuMG cells.
• Cells were fixed with PBS, 4% paraformaldehyde for 20 minutes at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT.
• After blocking, cells were incubated with diluted C1033-50E overnight at 4°C followed by Rhodamine Red-coupled anti-goat IgG at RT in the dark for one hour.
• Between each step, cells were washed with PBS, 0.1% BSA
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
• Reconstitute with sterile PBS.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
