The Janus family tyrosine kinases, consisting of Jak1, Jak2, Jak3 and Tyk2, associate with a variety of cytokine receptors and are activated by ligand binding to these receptors (1).
Upon ligand binding to cytokine receptors, Jaks phosphorylate themselves and their associated receptors, thereby providing multiple binding sites for signaling proteins containing SH2 or other phospho-tyrosine-binding domains, including Stats (2), Shc (3), insulin receptor substrates (4) and focal adhesion kinase (5).
The tyrosine residues 1022/1023 of Jak1 in the putative activation loop are the homologous tyrosine residues 1054/1055 in Tyk2, which are important in the regulation of Tyk2 kinase activity (6).
