In response to multiple cytokines and B and T cell receptor activation, the SH2 containing inositol 5' phosphatase, SHIP, becomes phosphorylated on tyrosine residues and associates with the tyrosine phosphorylated form of Shc.
It may serve as a negative regulatory molecule under certain conditions, by preventing the activation of mast cells as well as B cell expansion, and by promoting apoptosis of confluent hemopoietic cells.
Applications:Suitable for use in Immunoblot and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:Immunoblot Analysis: 0.5-2ug/ml Detects SHIP in RIPA lysates from human TF-1 cells (a bone marrow erythroleukemia cell line).
TF-1 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S1013-20A (2ug/ml).
Proteins were visualized using a goat anti-rabbit IgG labeled with HRP and a chemiluminescence detection system
Immunoprecipitation: 2ug Reported to immunoprecipitate SHIP from Ramos cells (human B cell line)
Optimal dilution determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
