Optineurin (OPTN) has recently been linked to glaucoma, a major cause of blindness worldwide.
Mutations in OPTN such as Glu50->Lys (E50K) have been reported in patients, particularly those with normal pressure glaucoma.
Apoptosis is controlled by a variety of signalling mechanisms and is relevant to several diseases and normal development.
Optineurin (originally named FIP-2) is a 74kD protein implicated in signal transduction of the tumor necrosis factor (TNF) pathway as presented in a study of adenovirus proteins that prevent cytolysis after TNF alpha stimulation (ref1).
Optineurin can be phosphorylated, however it is currently unknown what kinases achieve this or what other mammalian proteins interact with it downstream of TNF alpha or Fas signalling
Working at or near cell membranes and in concert with Rab8 and Huntingtin, optineurin has been implicated in membrane traffic regulation and cellular morphogenesis (ref 3).
The exact role of optineurin protein in apoptosis is currently unknown and interestingly optineurin gene mutations have been correlated with certain types of glaucoma (ref 4-6).
Increased apoptosis of trabecular meshwork cells is an identified factor for pathogenesis of glaucoma (ref 7).
Positive controls include murine or human heart, brain, or fibroblast lysates.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilutions:Western Blot: 1:200.
Incubate 1 hour at RT.
Overnight incubations at 4ºC at lower concentrations can produce optimal results

Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
