• Tau is a neuronal microtubule-associated protein found predominantly on axons.
• The function of tau is to promote tubulin polymerization and stabilize microtubules.
• Tau, in its hyperphosphorylated form, is the major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer’s disease (AD) brain.
• Hyperphosphorylated tau is also found in neurofibrillary lesions in a range of other central nervous system disorders.
• Hyper-phosphorylation impairs the microtubule binding function of tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons.
• Numerous serine/threonine kinases, including GSK-3b, protein kinase A (PKA), cyclin-dependent kinase 5 and casein kinase II, phosphorylate tau.
• Serine 356 can be phosphorylated by GSK-3b, PKA and MAP kinase and has been found to be a major site in AD brain.
• This antibody, combined with the tau [pS262] antibody, enables one to distinguish between phosphorylation at the two distinct residues, rather than a combined epitope as detected by 12E8 antibody.
• Applications:Suitable for use in Western Blot.
• Other applications have not been tested.
• Recommended Dilution:Western Blot: 1:1000
• Optimal dilutions to be determined by researcher.
• Positive Control:Recombinant human tau left untreated, or treated with PKA.
• Storage and Stability:May be stored at 4°C for short-term only.
• For long-term storage, aliquot and store at -20°C.
• Aliquots are stable for at least 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
