Tau is a neuronal microtubule-associatedprotein found predominantly on axons.
The function of tau is to promote tubulin polymerization and stabilize microtubules, but it also serves to link certain signaling pathways to the cytoskeleton.
Tau phosphorylation regulates both normal and pathological functions of this protein.
Tau, in its hyperphosphorylated form, is the major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer’s disease (AD) brain.
Hyperphosphorylation impairs the microtubule binding function of tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons.
Hyperphosphorylated tau is also found in a range of other central nervous system disorders.
Numerous serine/threonine kinases, including GSK-3b, PKA, PKC, CDK5,stress-activated protein kinases (SAPKs) and casein kinase II can phosphorylate tau.
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilution:Western Blot: 0.1-1ug/mlOptimal dilution to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage, store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
