Tau is a neuronal microtubule-associated protein found predominantly on axons.
The function of tau is to promote tubulin polymerization and stabilize microtubules.
Tau, in its hyper-phosphorylated form, is the major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer’s disease (AD) brain.
Hyper-phosphorylated tau is also found in neurofibrillary lesions in a range of other central nervous system disorders.
Hyper-phosphorylation impairs the microtubule binding function of tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons.
Numerous serine/threonine kinases, including GSK-3b, protein kinase A (PKA), cyclin-dependent kinase 5 (cdk5) and casein kinase II, hosphorylate tau.
Serines 199 and 202 are phosphorylated by GSK-3b and have been linked to hereditary frontotemporal dementia.
Serine 202 phosphorylation by cdk5, stimulated by the presence of microtubules, has been linked to hereditary neurodegenerative disease.
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilution:Western Blot: 1:1000Optimal dilution to be determined by the researcher.
Positive Control:Recombinant human Tau or African green monkey kidney (CV-1) cells, stably expressing human four repeat tau and SV40 small T antigen to induce specific inhibition of PP2A.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
