The Raf family of serine/threonine-specific kinases is comprised of three members (A-Raf, B-Raf and c-Raf) that play a critical role in regulating cell growth and differentiation and couple growth factor receptor stimulation to nuclear transcription factors via the Ras/mitogen-activated protein inase (MAPK) pathway.
c-Raf kinase (also known as Raf-1) is a key 74kD signal transducer of multiple extracellular stimuli that is regulated by several pathways and that once activated, phosphorylates MEK which in turn phosphorylates ERK.
Activation of Raf kinase activity is induced by several events including phosphorylation at several sites and binding of GTP-bound Ras and/or 14-3-3 proteins.
Integrin/growth factor receptor engagement induces phosphorylation of tyrosines 340 and 341 via Src, an event that is important in c-Raf activation.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000Optimal dilutions to be determined by the researcher.
Positive Control:HEK293 overexpressing c-Raf, in vitro-phosphorylated by active Src.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for at least 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
