Protein Kinase Cb (PKCb) is an 80kD member of the conventional group (cPKCs: sensitive to diacylglycerol, phosphotidylserine and phorbol esters) of the PKC family of serine/threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival, transcriptional regulation and tumor promotion.
PKCb has been implicated in diabetes and carcinogenesis.
PKCb isoforms I&II are phosphorylated on three sites, threonine 500 in the activation loop, bI threonine 642 (bII 641) in the turn loop and bI serine 661 (bII 660) in the hydrophobic loop.
Phosphorylation of PKCbI&II on threonine 500 by PDK1 is a prerequisite for its autophosphoylation and catalytic competence.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000 Optimal dilutions to be determined by the researcher.
Positive Control:K562 cells treated with PMA, a phorbol ester
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for at least 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
