The Lin-11, Is1-1 and Mec-3 kinases 1 and 2 (LIMK1 [72kD] and LIMK2 [65kD]) are serine/threonine kinases that are involved in actin cytoskeletal reorganization, cell growthregulation and tumor supression.
LIMK1/2 are activated by various extracellular stimuli including stromal cell-derived factor-1alpha (SDF-1alpha) and insulin, through activation of small GTPases (Rho, Rac-1 and Cdc-42) and their downstream kinases, ROCK and PAK1.
ROCK directly phosphorylates LIMK1 at threonine 508 and LIMK2 at threonine 505.
LIMK1 plays a role in Racinduced lamellipodia formation, whereas LIMK2 plays a role in Cdc42-induced filipodial formation and Rho-induced stress fiber formation.
LIMK1/2 negatively regulate cofilin activity by phosphorylating cofilin at serine 3 and hindering it from associating with actin.
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000Optimal dilutions to be determined by the researcher.
Positive Controls:Jurkat cells treated with SDF-1alpha; A431 cells treated with EGF.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage, store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
