Inhibitor of NFkB a (IkBa) is a ~40kD protein that has a central role in the regulation of transcription mediated by the NFkB transcription factor.
In the absence of a stimulus, NFkB is sequestered in the cytoplasm through binding to IkB proteins.
A variety of stimuli (e.g., proinflammatory cytokines, Tumor Necrosis Factor-a, and interleukin-1b) lead to phosphorylation and activation of the IKK complex (consisting of IKKa, b, and y).
The activated IKK complex in turn phosphorylates IkBa serines 32 and 36, leading to its subsequent ubiquitination and degradation by the proteasome.
The free NFkB translocates to the nucleus and stimulates transcription of NFkB-responsive genes.
Phosphorylation of these serine residues by several other kinases, including p90S6 kinase (Rsk1), NFkB-Inducing Kinase (NIK), and Mitogen-Activated Protein Kinase Kinase Kinase-1 (MEKK1), with subsequent activation of NFkB-mediated transcription, has also been reported.
A component of the NFkB complex, RelA, has recently been shown to regulate the level of IkBa phosphorylation.

Recommended Dilution:Western Blot: 1:1000 Optimal dilutions to be determined by the researcher.
Recommended Positive Control:Jurkat cells pretreated for 30 min.
with N-acetly-leucinyl-leucinyl-norleucinal (ALLN) and stimulated with TNF-alpha.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
