Focal Adhesion Kinase (FAK) is a 125kD non-receptor protein tyrosine kinase that plays a key role in signaling by growth factors, extracellular matrix and stress signals.
Indeed, FAK plays a central role in cell spreading, differentiation, migration, cell death and acceleration of the G1 to S phase transition of the cell cycle.
The serine/threonine kinase cyclin-dependent kinase-5 (Cdk5)-an important regulator in brain development-phosphorylates serine 732 of FAK, possibly regulating a centrosome-associated microtubule structure to promote nuclear translocation, a critical step in neuronal migration.
This phosphorylation does not have a direct impact on the kinase activity of FAK, but appears to prevent the accumulation of FAK at the centrosome.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000 Optimal dilutions to be determined by the researcher.
Positive Control:PC12 cells treated with nocozadole; cdk5 +/- and cdk5 -/- mouse brain lysates.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for at least 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
