b-catenin (92kD) is a multifunctional protein involved in cell-cell adhesion, intracellularsignaling and gene transcription.
b-catenin plays a key role in the Wnt pathway which mediates numerous developmental processes.
Deregulation of the Wnt pathway results in the accumulation of b-catenin in the nucleus.
Stabilizing mutations of several serine/threonine residues (more frequently of serine 45) have been found in multiple tumors.
Phosphorylation of b-catenin on serine 45 by casein kinase I (CKI) primes its subsequent phosphorylation of residues 33, 37 and 41 by glycogen synthase-3b (GSK-3b) and results in its degradation.
