The tumor suppressor protein, p53, is expressed ubiquitously and plays an important role in blocking proliferation of damaged cells.
Phosphorylation of p53 at serine 392 is reported in response to diverse stresses, for example UV irradiation.
The specificity of this antibody has been shown by blocking studies in Western blotting, whereby staining was blocked by the phosphopeptide.
No blocking was seen by the corresponding non-phosphopeptide.
Applications:Suitable for use in Western Blot and Immunohistochemistry (frozen).
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000, detects bands of ~53kD in in rat brain nuclear fraction lysates.
For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM sodium fluoride should be added to the sample buffer.
Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background
Optimal dilution determined by the researcher.
Recommended Secondary Reagents:Sheep anti rabbit IgG:HRP
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
