Dynamin is a phosphoprotein expressed at nerve terminals, which plays an important role in the regulation of synaptic vesicle endocytosis.
Cyclin dependent protein kinase 5 (cdk5) has been shown to phosphorylate serine 774 and 778 in vivo.
Dephosphorylation of these residues is essential for endocytosis of synaptic vesicles.
Applications:Suitable for use in Western Blot, Immunofluorescence and Immunohistochemistry (frozen).
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000, detects a band of ~95kD in rat hippocampus lysates.
For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM sodium fluoride should be added to the sample buffer.
Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background
Optimal dilution determined by the researcher.
Recommended Secondary Reagents:Donkey anti sheep/goat IgG:HRP
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
