Tight junctions create circumferential seals around cells in epithelial tissues that are critical for maintaining cell-cell adhesion.
The “barrier” function of tight junctions regulates the passage of ions, water, and various macromolecules through paracellular spaces, while the “fence” function maintains cell polarity.
Tight junctions thus function as a fence to prevent intermixing of molecules in the apical membrane with those in the lateral membrane.
Tight junctions are comprised of the membrane proteins occludin, claudins, and JAMs.
Claudins are responsible for the formation of tight junction strands and are connected with the actin cytoskeleton by ZO-1
Claudin-15 is expressed in endothelial cells in the mouse kidney.
Claudin-15 creates Na+ -selective paracellular channels, and charge-reversing mutations on the first extracellular domain of Claudin-15 create Cl- -selective paracellular channels.
Claudin-15 has a high level of expression in the mouse small and large intestine.
