The 90kD ribosomal S6 kinases (RSK1-3) are a family of broadly expressed serine/ threonine kinases activated by extracellular signal-regulated protein kinases (Erk1 and 2) in response to many growth factors, polypeptide hormones and neurotransmitters.
p90RSK (PSK1) lies immediately downstream of MAP kinase in the phorbol ester and growth factor singaling pathway (1,2).
PSK1 is activated by MAPK in vitro and in vivo via phosphorylation (3).
Several phosphorylation sites, including Ser380, Thr359 and Ser363, are important for its activation (4,5).
RSK3 resembles PSK1 by having two kinase domains connected by a regulator linker region.
Phosphorylation of PSK1 (and likely RSK2 and RSK3) in the linker region is critical for its activity.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
