Herstatin (Hst), a secreted HER-2 gene product, binds to the HER-2 receptor ectodomain blocking receptor activation.
Herstatin is an autoinhibitor of the ErbB family consisting of subdomains I and II of the human epidermal growth factor receptor 2 (ErbB-2), extracellular domain and a novel C-terminal domain encoded by an intron.
Herstatin binds to human epidermal growth factor receptor 2 and to the epidermal growth factor receptor (EGFR), blocking receptor oligomerization and tyrosine phosphorylation.
Applications:Suitable for use in ELISA and Western blotting.
Titer: Defined by indirect ELISA is >1:100,000 for antibody concentration 1mg/ml, 25ng of antigen are coated per well, and is then defined at a point of maximal decrease of the titration curve.
Reconstitution: Add 0.05ml of deionized water and let the lyophilized pellet dissolvecompletely.
Slight turbidity may occur after reconstitution, which does not affectactivity of the antibody.
In this case clarify the solution by centrifugation.
Storage and Stability: May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Freeze at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
