Adiponectin (also called ACRP30 and GBP28) is a protein exclusively secreted from adipose tissue.
Full-length adiponectin was expressed in HEK293 cells stably transfected with pcdm-F vector encoding FLAG epitope-tagged mouse adiponectin.
Recombinant adiponectin was purified using anti-FLAG M2 affinity gel (Sigma-Aldrich) and was eluted with FLAG peptide.
FLAG peptides were removed by an extensive wash with an excessive volume of saline.
Highly sensitive matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass-spectrometric analysis confirmed that FLAG peptide was not detectable in the protein solution.
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:Indirect ELISA: >1:100,000 using 25ng of antigen coated per well
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
