Non-insulin-dependent diabetes mellitus (NIDDM) is a multifactoral disease with both environmental and genetics causes.
Genome-wide screening procedures have identified several susceptibility loci for NIDDM within the human genome.
A putative sugar transporter that has been localized to human chromosome 20q12-q13.1, one of the genomic loci associated with NIDDM.
Because of the strong resemblance of this novel protein to members of the mammalian facilitative glucose transporter family (GLUT), the protein is known as GLUT10 (HGMW-approved gene symbol SLC2A10).
Data suggests that GLUT10 an excellent candidate for a susceptibility gene involved in NIDDM.

NCI-H1299 non-small cell lung tumor cells were transfected with the eukaryotic expression vectors pcDNA3.1/V5 (lanes 1,2), pCMV-GLUT10 (lanes 3,4), and pCMV-GLUT1 (lanes 5,6).
Two days after transfection total cell extracts were prepared and protein content quantitated with the BCA assay.
For Western blot analysis, 30ug of protein were loaded onto 10% polyacrylamide-SDS gels, transferred to ECL-grade nitrocellulose membranes, and incubated with a 1:100 dilution of the anti-GLUT10 antibody for 2 h at room temperature.
For detection with the ECL system (Amersham), a 1:10000 dilution of the HRP-conjugated sheep anti-rabbit secondary antibody (Amersham) was employed for 1 h at room temperature.
Upon the addition of the ECL detection reagents x-ray film was exposed for 10 min.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Store at -20°C or colder.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
