It combines many of the benefits of E.
coli expression with the advantages of expression in an eukaryotic system such as protein processing, folding, and posttranslational modifications.
Pichia pastoris is capable of metabolising methanol as the sole carbon source.
The first step in the metabolism of methanol is the oxidation of methanol to formaldehyde by the enzyme, alcohol oxidase.
Expression of this enzyme, coded for by the AOX1 gene, is tightly regulated and induced by methanol to very high levels:- typically >30% of the total soluble protein in the cells.
Applications:Suitable for use in Immunofluorescence, ELISA, Western Blot and Immunocytochemistry.
It is also suitable for conjugation purposes.
Other applications not tested.
Recommended Dilution:Western Blot: >1:100Immunofluorescence: 1:800Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
