The family of serine/threonine protein kinases Akt (also called protein kinase B or PKB) plays a key role in a variety of biological processes including cell survival, cell growth, gene expression, and oncogenesis.
Upon cell stimulation, the kinase is translocated to the plasma membrane, where it is phosphorylated on two amino acids, Thr 308 in the catalytic domain and Ser 473 in the COOH-terminal regulatory domain.
The activation/phosphorylation of AKT/PKB is essential in its cellular signal transduction.
Applications:Suitable for use in ELISA, Immunoprecipitation and Western Blot.
Other applications not tested.
Recommended Dilution:ELISA: 0.1-1.0ug/mlImmunoprecipitation: Use 3-5ug for 10e6 cells
Western Blot: 0.5-2.0ug/ml; Predicted molecular weight: 56kD.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Freeze at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
