• Several subtypes of NPY Receptors have been described of which the cloned Y1-, Y2- and Y5-receptors, members of the superfamily of G-protein coupled receptors, appear to be the endogenous binding sites for NPY.
• The Y(2) receptor is the predominant Neuropeptide Y (NPY) receptor subtype in the brain.
• Y(2) receptor-mediated effects have been reported principally based on the observations using the C-terminal fragments of NPY.
• Recent studies indicate an involvement of the receptor in food intake, gastrointestinal motility, cardiovascular regulation, and neuronal excitability.
• Applications:Suitable for use in ELISA, Immunohistochemistry and Immunofluorescence.
• Other applications not tested.
• Recommended Dilutions:Immunohistochemistry: 1:500-1:1000; Used on perfusion fixed tissue.
• Perfusion: 1) calcium-free Tyrode’s solution, 2) paraformaldehyde-picric acid fixative and 3) 10% sucrose in PBS.
• Tissues were dissected and stored overnight in 10% sucrose, PBS
• Immunofluorescence (Indirect): Incubate slides with blocking buffer for 1 hour at RT.
• Dilute N2176-63G with blocking buffer.
• Remove blocking buffer from slide and incubate with N2176-63G for 18-24 hours at 4°C.
• Rinse slides 3X and then incubate with appropriate secondary antibody for 1 hour at RT.
• Rinse slides again 3X and set coverslip.
• Examine staining using fluorescence microscopy
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
