特徴

• Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells.
• Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure.
• The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes.
• These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.
• Applications:Suitable for use in Immunofluorescence, Western Blot and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilution:Western Blot: 1:5000-1:10,000 detects phosphorylated Histone H3 in acid extracted proteins from mitotic HeLa cells but not unphosphorylated recombinant Histone H3.
• Recombinant Histone H3 protein and acid extract from colcemid-treated HeLa cells were resolved by electrophoresis, transferred to nitrocellulose and probed with a 1:5,000 dilution of antiphospho-Histone H3 (Thr3).
• Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system
• Optimal dilutions to be determined by the researcher.
• Positive Control:Acid extract from colcemid-treated HeLa cells
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
