• SGK1 is a member of the serum- and glucocorticoid-induced protein kinase family.
• SGK1 is activated in vitro by 3-phosphoinositide- dependent protein kinase-1 (PDK1) and in vivo in response to signals that activate phosphatidylinositol (PI) 3-kinase.
• SGK1, mRNA is expressed in all tissues and the level of SGK1 mRNA is increased by stimulation with serum or dexamethasone.
• Applications:Suitable for use in Western Blot.
• Other applications have not been tested.
• Recommended Dilutions:Western Blot: 1:1000-1:3000.
• Not suitable for use with cell lysates.
• Detects phospho- SGK1 after phosphorylation of unactive SGK1 (S1010-82) with PDK1.
• Unactive SGK1 was incubated with or without PDK1 for 30 min at 30ºC.
• Phosphorylation was termiated by boiling in reducing sample buffer.
• Aliquots (50ng) were resolved by electrophoresis, transferred to nitrocellulose and probed with S1010-84 (1:3000).
• Proteins were visualized using an IgG (HRP) goat anti rabbit secondary antibody and a chemiluminescence detection system
• Optimal dilutions to be determined by the researcher.
