PGP9.5/UCH-L1 is a member of a gene family whose products hydrolyze small C-terminal adducts of ubiquitin to generate the ubiquitin monomer.
Protein gene product (PGP9.5) is a neuron specific protein, structurally and immunologically distinct from neuron specific enolase.
The protein which has a molecular weight of 27kD was first identified by high resolution two dimensional PAGE.
Standard immunohistochemical techniques have demonstrated the presence of PGP9.5 in neurons and nerve fibres at all levels of the central and peripheral nervous system, in many neuroendocrine cells, in segments of the renal tubules, in spermatogonia and Leydig cells of the testis, in ova and in some cells of both the pregnant and non-pregnant corpus luteum.
PGP9.5 gene encodes two opposing enzymatic activities that affect alpha-synuclein degradation and Parkinson's disease susceptibility.
Applications:Suitable for use in Immunohistochemistry.
Perfusion: 1) calcium-free Tyrode’s solution, 2) paraformaldehyde-picric acid fixative, and 3) 10% sucrose in PBS as a cryo-protectant.
Desired tissues were dissected and stored overnight in 10% sucrose in PBS.
Slide-mounted tissue sections were processed for indirect immunofluorescence.
Slides were incubated with blocking buffer for 1 hour at room temperature.
P9102-71C was diluted with blocking buffer to the appropriate working concentration.
Blocking buffer was removed and slides were incubated for 18-24 hours at 4ºC with P9102-71C.
Slides were rinsed 3 times and then incubated with secondary antibodies for 1 hour at room temperature.
Slides were again rinsed 3 times and coverslipped.
Staining was examined using fluorescence microscopy
Note: Sodium azide interferes with peroxidase reactions and should not be used with peroxidase methodologies.
If sodium azide is present in any steps of the staining procedure, the tissue should thoroughly be rinsed with sodium azide-free buffer before performing the peroxidase reaction.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
