Parkinson’s disease, a common neurodegenerative disease is caused by slow death of neurons in the substantial nigra, a brain region that utilizes the neurotransmitter dopamine.
Parkin, a recently discovered gene encoding a large protein, may be involved in normal and abnormal protein degradation in cells.
Recent evidence indicates that point mutations in the Parkin gene appears to be responsible for the pathogenesis of some forms of Parkinson’s disease.
The Parkin protein is expressed in various regions of the brain, including the substantia nigra, and is also expressed in other tissues, including heart, testes and skeletal muscle.
This protein may function in the control of cell growth, differentiation and development.
Parkin is a protein of 465 amino acids with an amino-terminal ubiquitin domain and a carboxy-terminal RING-box (1).
In the case of autosomal recessive juvenile Parkinsonism (AR-JP), deletions have been found in the gene on chromosome 6 encoding the protein Parkin.
Applications:Suitable for use in ELISA, Western Blotting.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000-1:5000 for neat serum and 1-10ug/ml for affinity pure using ECL
ELISA: ~1:100,000 using 50-100ng control peptide/well.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Freeze at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.

Specificity:Human peptide sequence is conserved in alternatively spliced isoforms (isoform 1, 465 aa; isoforms 2, 437 aa; and isoform 3, 316 aa;;). It is also highly conserved in mouse and rat (95%) and drosophila's Parkin (69%). No significant homology is seen with ubiquitin or any other proteins. Antibody crossreactivity with parkin from other species is not known. Control peptide, because of its low MW (<3kD), is not suitable for Western. It should be used for ELISA or antibody blocking experiments (use 5-10 ug control peptide per 1 ug of aff pure IgG or 1 ul antiserum) to confirm antibody specificity
