• Na+/H+ exchangers (NHE) of mammalian cells are plasmamembrane intrinsic proteins mediating exchange of N+ and H+ ions in various tissues.
• The NHE catalyzes the electroneuraltransport of extracellular Na+ for intracellular H+.
• They play amajor role in regulation of intracellular pH (pHi) in addition totrans-cellular absorption of Na+, cell volume regulation andpossibly in cell proliferation.
• These primary functions of theNa+/H+ exchanger have been related to many pathophysiological states, include hypertension, organ growthand hypertrophy, regression of cancer and renal intestinaldisorders.
• Five NHE isoforms (NHE1-5) have been cloned sofar.
• They are all similar in their primary structure and predictedto have 10-12 transmembrane domains.
• The COOH-terminalsof NHE1, NHE2 and NHE3 are intracellular.
• NHE1 (rat/mouse; 820aa; human 815aa; chromosome 1 p35-36.1) is 'House Keeper' and is expressed ubiquitously in all tissues.
• It is implicated in pH homeostasis, volume regulation and probably cell proliferation.
• It is expressed on basolateral surface of several epithelia (1).
• Applications:Suitable for use in ELISA.
• Western Blot, though not tested, may potentially be used as an application.
• Other applications not tested.
• Recommended Dilution:ELISA: 1:100,000 using 50-100ng N2015-09 Control peptide/wellWestern Blot: 1:1000-1:5000 using Chemiluminescence technique.
• A band of 95-110kD has been detected in brush border membrane (2)
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for at least 12 months.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
