Escherichia coli MutY is an adenine DNA glycosylase active on DNA substrates containing A/G, A/8-oxoG, or A/C mismatches and also has a weak guanine glycosylase activity on G/8-oxoG-containing DNA.
The N-terminal domain of MutY, residues 1-226, has been shown to retain catalytic activity.
The post-replicative adenine:8-oxo-7,8-dihydroguanine (GO) mismatch is crucial for G:C to T:A transversion.
This mismatch is corrected by Escherichia coli MutY which excises the adenine from A:GO.
A candidate gene coding for the human counterpart of MutY has been cloned as hMYH (535 aa) (1).
Human MutY is approximately 55kD.
Applications:Suitable for use in ELISA.
Western Blot, though not tested, may potentially be used as an application.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500-1:2000
ELISA: 1:5000-20,000 using 50-100ng control peptide (M9559-01)/well.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage, store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
