Recommended Dilution:Western Blot: 1:1000-1:5000 using neat serum or 1-10ug/ml for affinity Pure Antibodies) with Chemiluminescence technique.
The obese protein is predicted to be secretory protein with a molecular weight of ~ 16kD (refs.
1, 2).
ELISA: 1:10,000-50,000 10-100ng of control peptide or Leptin protein/well).
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%).
Freeze at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.

Specificity:The specificity of the anti-Ob12 has been tested by control Ob2 and leptin in an ELISA. The antibodies reacted only with their respective peptides. The peptide sequences is the same in mouse and human Ob proteins and the antibodies should be crossreactive. Anti-Ob12 has detected recombinant mouse leptin and human leptin in both native samples by dot blot and denatured and reduced samples by Western Blots (sensitivity of about 1-5 ng by Chemiluminescent detection). Control peptide, because of its small size (2-3kD), is not recommended for Western. It is recommed for ELISA, dot blot, or for antibody blocking experiments (use 5-10 ug of control peptide per 1 ug of aff pure IgG or 1 ul of antiserum) to demonstrate antibody specificity
