• Tyrosine hydroxylase catalyzes the rate limiting step in the synthesis of dopamine and other catecholamines.
• The enzyme’s activity is regulated at the levels of transcription, translation and posttranslational modification (1,2).
• Tyrosine hydroxylase exists in several different isoforms ranging from 50kD to about 62kD in human.
• The N-terminal regulatory domain contains three phospho-serine sites: serine 19, serine 31 and serine 40 (4,5).
• Phosphorylation at serine 40 by PKA positively regulates the enzyme catalytic activity of the enzyme (4-6).
• Applications:Suitable for use in ELISA and Western Blot.
• Other applications not tested.
• Recommended Dilution:Western Blot: 1:1000, incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
