PTEN (phosphatase and tensin homologue deleted on chromosome ten)/MMAC (mutated in multiple advanced cancers) phosphatase is a tumor suppressor gene implicated in a wide variety of human cancers (1).
PTEN encodes the 403 amino acid polypeptide originally described as a dual-specificity protein phosphatase (2).
The main substrates of PTEN are inositol phospholipids generated by the activation of the phosphoinositide 3 kinase (PI3K) (3).
PTEN is a major negative regulator of the PI3K/Akt signaling pathway (1,4–6).
PTEN possesses a C-terminal noncatalytic regulatory domain that contains three phosphorylation sites (Ser380, Thr382 and Thr383), which regulates its stability and could play an important role in control of its biological activity (7,8).
Applications:Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight
Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
